UspA belongs to a superfamily of widely conserved proteins whose physiological function is unknown. Synthesis of UspA is induced in response to a variety of stresses [Nystrom92]. UspA protein is autophosphorylated at serine/threonine residues in response to the same conditions that induce its expression [Freestone97]. The overlapping and distinct roles of the various Usp proteins in E. coli have been studied [Nachin05].

A uspA null mutant has a metabolic defect, leading to a diauxic growth phenotype when diluted into fresh medium [Nystrom93]. Mutants also show a defect in survival of prolonged periods of growth inhibition [Nystrom94a] and are sensitive to UV irradiation and mitomycin C exposure [Diez00]. Overexpression of UspA leads to decreased growth rate in minimal medium and causes global changes in protein synthesis [Nystrom96].

uspA mRNA stability during glucose starvation is diminished in the absence of RNase III function [Freire06].

Transcription of uspA is repressed by FadR [Farewell96], but repression can be overcome by the alarmone ppGpp during severe cellular stress [Kvint00].
