MtfA interacts with the transcriptional repressor Mlc, thereby modulating the expression of genes which are regulated by Mlc [Becker06]. Binding of MtfA to Mlc prevents interaction of Mlc with the ptsG operator region [Gohler12].
The C-terminal region of MtfA is responsible for interaction with Mlc [Gohler12].
MtfA has aminopeptidase activity. The Mlc-binding and peptidase activities of MtfA are separable, and Mlc does not appear to be a biologically relevant peptidase substrate [Gohler12].
An mtfA insertion mutant shows slow growth on glucose and decreased expression of ptsG, the gene encoding the major glucose transporter in the cell. Expression of ptsG is regulated by several mechanisms, including transcriptional repression by Mlc. Mlc is epistatic to MtfA, and MtfA interacts with Mlc directly in vitro [Becker06].
