Summary:
The transcription factor CysB, for "Cysteine B," is negatively autoregulated [JaguraBurdzy81, Jovanovic03]; this regulator also controls the transcription of the operon involved in novobiocin resistance [Lilic03, Jovanovic03] and transcription of genes involved in sulfur utilization and sulfonate-sulfur catabolism via cysteine biosynthesis [JaguraBurdzy81, vanderPloeg97, Monroe90, Lochowska01, Lochowska04, Kredich92, Leyh88, vanderPloeg01, Rakonjac91]. In the presense of N-acetylserine, the inducer molecule of this regulator, DNA binding by CysB tetramers to the target sequences is facilitated [Lochowska01, Jovanovic03], but the binding to the repressor site in the cysB regulatory region is prevented [Ostrowski91].

The amino acid sequence of CysB shows homology to members of the LysR family of transcriptional regulators [Tei90, Ostrowski87, Tyrrell97, Jovanovic03]. The N-terminal region of this protein comprises a helix-turn-helix DNA-binding motif. CysB has a central region, suggesting that it may be important for inducer recognition [Tyrrell97], and the carboxy-terminal domain is involved in homo-oligomerization, stability, and DNA binding [Lamark91, Lochowska01]. Tyrrel et al. proposed that this regulator has an unusual mechanism of binding to DNA [Tyrrell97].
