Summary:
MdtK is a putative multidrug transporter and probably functions as a drug/proton antiporter. MdtK is a member of the MATE (multidrug and toxic compound extrusion) family of multidrug efflux transporters [Brown99] and is homologous to NorM of Neisseria gonorrhoeae [Long08].

Expression of the cloned mdtK gene conferred increased resistance to norfloxacin, ciprofloxacin, acriflavine, and tetraphenylphosphonium [Morita98]. E. coli strains expressing norM or mdtK showed elevated resistance to several antimicrobial agents, including fluoroquinolones, ethidium bromide, rhodamine 6G, acriflavine, crystal violet, berberine, doxorubicin, novobiocin, enoxacin, and tetraphenylphosphonium chloride. Steady-state fluorescence polarization assays reveal that MdtK binds to these antimicrobials with dissociation constants in the micromolar region [Long08].

Overexpression of ydhE rescues the hypermutable phenotype of an 8-oxoguanine repair deficient E.coli strain possibly by extrusion of molecules that oxidise deoxy-guanine [Guelfo10].
