Summary:
TldD and/or TldE act as a protease [Allali02] that is involved in sensitivity to DNA gyrase inhibitor LetD/CcdB (encoded by the F plasmid) [Murayama96] by effecting degradation of the F plasmid-encoded LetD inhibitor, LetA/CcdA [Allali02]. TldD [Allali02] and TldE [RodriguezSainz90, Allali02] are also involved in proteolytic processing of the antibiotic Microcin B17 (encoded by the pMccB17 plasmid) prior to its export from the cell. TldD, TldE and GroE have been proposed to antagonize ZfiA-mediated inhibition of the binding of the F-plasmid-encoded LetD DNA gyrase inhibitor to the DNA gyrase A subunit [Murayama96].

A tldD [Allali02] or tldE [RodriguezSainz90, Allali02] mutant harboring the pMccB17 episome exhibits accumulation of Microcin B17 precursor protein, in contrast to wild type, which exhibits processing and the export of Microcin B17 from the cell. A tldD, tldE, or groES mutant exhibits resistance to the F-plasmid-encoded LetD DNA gyrase inhibitor [Murayama96]. A zfiA mutation suppresses the resistance to the F-plasmid-encoded LetD DNA gyrase inhibitor exhibited by a tldD, tldE or groES mutant [Murayama96].

TldE is cytoplasmic [RodriguezSainz90].
