Escherichia coli K-12 substr. MG1655 Transporter: heavy metal divalent cation transporter ZupT

Gene: zupT Accession Numbers: EG11167 (EcoCyc), b3040, ECK3031

Synonyms: ygiE

Regulation Summary Diagram: ?

Summary:
ZupT is a member of the ZIP family of divalent metal cation transporters, and is predicted to have 8 transmembrane domains. Transporter assays along with mutation and complementation studies indicate that ZupT transports zinc, copper [Grass02], cadmium [Taudte10] iron, cobalt, and manganese into the cell [Grass05]. ZupT is expressed constitutively at a low level as a monocistronic transcript [Grass05].

The energetics of ZupT transport are not clear: metal uptake is inhibited by the ionophores FCCP (carbonyl cyanide-p-trifluoromethoxyphenyl hydrazone) and CCCP (carbonyl cyanide m-chlorophenyl hydrazone) - suggestive of a role for proton motive force - but metal uptake at pH 7.2 is higher than that at pH 6 or pH 6.5, which argues against a proton/metal symport mechanism [Taudte10].
