A yghB yqjA double null strain has a synthetic growth defect at elevated temperatures, forms chains of cells at permissive temperatures, and can be complemented by yghB, yqjA, yohD, or yabI expressed from a plasmid [Thompkins08]. The double and single mutants displayed altered phospholipid compositions [Thompkins08].
An E. coli ΔyqjA ΔyghB strain is hypersensitive to a number of structurally diverse drugs and dyes including ethidium bromide, methyl viologen, acriflavine, nalidixic acid and β-lactam antibiotics. A wild-type response is restored by expression of yqjA from a plasmid [Kumar14]. Mutation of the membrane embedded acidic amino acids E39 (glutamic acid) or D51 (aspartic acid) in plasmid-expressed YqjA compromises its ability to restore wild-type resistance [Kumar14]. Overexpression of yqjA in a wild-type background does not result in increased resistance to any of the compounds tested. Overexpression of the mdfA antiporter restores the wild-type resistance of an E. coli ΔyqjA ΔyghB strain to all componds tested as does growth in the presence of 400mM NaCl or with increased amounts of cations or at pH 6 [Kumar14, Thompkins08]. Deletion of yqjA and yghB may indirectly cause drug sensitivity due to impairment of the proton-motive force (PMF) [Kumar14].
YqjA is a member of the DedA family of inner membrane proteins [Thompkins08, Doerrler13]. E. coli K-12 contains 7 other DedA family members, namely DedA, YghB, YabI, YohD, YqaA, YdjX and YdjZ. Collectively, the DedA family is essential in E. coli K-12 [Boughner12].
