Summary:
Spy is an ATP-independent periplasmic chaperone that prevents protein aggregation and assists protein refolding.

Purified Spy reduces the aggregation of thermally denatured malate dehydrogenase and protects DsbB and alkaline phosphatase from tannic acid induced aggregation [Quan11]. Purified Spy increases the refolding of malate dehydrogenase and lactate dehydrogenase as assessed by enzymatic activity; refolding occurs in the absence of any cofactors [Quan11]. Spy sediments upon ultracentrifugation as a 30kDa species suggesting that it forms dimers in solution [Quan11].

Expression of spy is strongly induced by the protein denaturants butanol, ethanol and tannic acid [Quan11]. spy is also strongly induced in baeS constitutive mutants [Quan11]. spy null mutants are highly sensitive to tannic acid and show decreased alkaline phosphatase activity in vivo [Quan11]. The production of Spy is induced by spheroplasting and further increased in the presence of glucose [Hagenmaier97]. Expression of spy is regulated by the CpxAR two-component signal transduction system and by the BaeSR two-component system [Raivio00, Raffa02].

A crystal structure of Spy from E.coli O157:H7 has been resolved at 2.6 Å [Quan11]

Secretion of Spy is dependent on the the Sec pathway for protein export [Hagenmaier97].
