The subunit structure of this enzyme has not been reported.

Streptomyces sp. WA46 was isolated from soil and shown to degrade salicylate via a novel pathway involving its coenzyme A derivative (see pathway salicylate degradation IV). A plasmid containing a gene cluster sdgABCDEFGHR from Streptomyces sp. WA46 was constructed and shown to be sufficient for the complete degradation of salicylate. Derivatives disrupted in each gene were created and transformed into the heterologous host Streptomyces lividans TK64. salicylate degradation was blocked by disruption of genes sdgA and sdgC and gentisate accumulation resulted from disruption of sdgD [Ishiyama04].

Expression of individual genes in Escherichia coli and assay of enzyme activities in cell extracts suggested their function. Gene sdgA was required for the conversion of salicylate to salicyloyl-CoA with cofactors ATP and coenzyme A via a postulated salicylate-adenylate intermediate. The product of gene sdgB had amino acid sequence similarity to acetyl-CoA synthetases. It was hypothesized to be involved in coenzyme A addition [Ishiyama04].

The product of gene sdgB was proposed to be involved in coenzyme A addition in this reaction although this was not demonstrated. salicyloyl-CoA was produced by SdgA in the presence or absence of SdgB [Ishiyama04].
