MetaCyc Enzyme: ent-kaurene synthase

Summary:
A cDNA clone was isolated from the moss Physcomitrella patens and recombinantly expressed and characterized. The expressed protein is a bifunctional enzyme with similarity to the fungal bifunctional copalyl diphosphate synthase (CPS) and ent-kaurene synthase (KS). This enzyme catalyzes the conversion of geranylgeranyl diphosphate into ent-16-a-hydroxy-kaurene without the formation of the intermediate copalyl diphosphate as observed in higher plants. By mutation complementation studies, it was shown that the enzyme sequence has the signature motifs DVDD and DDYFD. The DVDD motif has the CPS actvity and the DDYFD has the KS activity [Hayashi06a].
