Summary:
A cDNA clone was isolated from wheat germ. The cDNA ecodes E1 an ubiquitin-activating enzyme. The enzyme sequence had 1051 amino acids. Heterologous expresseion and characterization of the recombianant protein UBA 1 showed the product of UBA1 catalyzed the formation of a thiol ester linkage between ubiquitin and an ubiquitin carrier protein [Hatfield90].

a [ubiquitin-activating protein E1]-L-cysteine + a [ubiquitin] C-terminal glycine + ATP <=> an S-ubiquitinyl-[ubiquitin-activating protein E1]-L-cysteine + AMP + diphosphate
