Summary:
DesI belongs to the aspartate aminotransferase family, and catalyzes the PLP-dependent amination of the C-4' position in dTDP-4-dehydro-6-deoxy-α-D-glucopyranose, the first committed step in the biosynthesis of dTDP-D-desosamine. This ping-pong reaction first requires the conversion of PLP to PMP using glutamate as the nitrogen source, followed by amination of the sugar substrate.

The desI gene, which encodes the enzyme, was cloned from Streptomyces venezuelae and expressed in Escherichia coli, and the recombinant protein was purified to homogeneity [Burgie07]. The structure of DesI was solved in complex with its product, dTDP-viosamine, to a nominal resolution of 2.1A [Burgie07].
