Summary:
The Xanthobacter autotrophicus Py2 alkene monooxygenase (Xamo) catalyzes the epoxidation of a range of alkenes, some with a high degree of stereospecificity, but does not hydroxylate the homologous alkanes [vanGinkel87].

The enzyme has been resolved into four components: an NADH-dependent reductase, a Rieske-type ferredoxin, an oxygenase, and a small protein which may be a coupling or effector protein. The oxygenase component is a hexameric comlex composed of three types of polypeptides in an α2β2γ2 structure [Small97].

The six genes encoding the enzyme components have been located and analyzed. Sequence analysis revealed that the enzyme is highly homologous to aromatic monooxygenases, such as toluene-4-monooxygenase| [Zhou98b]. Not surprisingly, the enzyme is capable of oxidizing benzene, phenol and toluene [Zhou99].

Enzymatic reaction of: propylene monooxygenase (alkene monooxygenase)

propylene + NADH + oxygen + H+ <=> (R)-1,2-epoxypropane + NAD+ + H2O

The reaction direction shown, that is, A + B ↔ C + D versus C + D ↔ A + B, is in accordance with the Enzyme Commission system.

The reaction is physiologically favored in the direction shown.

Alternative Substrates for propylene: benzene [Zhou99 ] , phenol [Zhou99 ] , toluene [Zhou99 ] , trichloroethylene [Zhou99 ] , ethylene [Zhou99 ] , butylene [Zhou99 ] , 1,3-dichloropropene [Zhou99 ] , vinyl-chloride [Zhou99 ]

In Pathways: propylene degradation

Cofactors or Prosthetic Groups: diiron [Small97]

Enzymatic reaction of: propylene,NADH:oxygen oxidoreductase (alkene monooxygenase)

propylene + NADH + oxygen + H+ <=> (S)-1,2-epoxypropane + NAD+ + H2O

The reaction direction shown, that is, A + B ↔ C + D versus C + D ↔ A + B, is in accordance with the Enzyme Commission system.

The reaction is physiologically favored in the direction shown.

In Pathways: propylene degradation

Cofactors or Prosthetic Groups: diiron [Small97]
